研究概要

Relは、免疫細胞の機能発現に不可欠であり、ヒトの疾患とも関連が深い。本研究は、Relの活性化機構及びその生物学的意義を明らかにすることを目的とする。
1.Rel/1kB複合体を活性化するシグナルの解析:
活性化シグナルの伝達因子を同定するため、酵母ツ-ハイブリット法によりCD40細胞質領域に結合する二種類の新規TRAF蛋白質(TRAF5,TRAF6)のcDNAを得た。どちらも過剰発現によりRel/1kB複合体を活性化した。CD40変異体を用いた実験から、TRAF5の結合領域はアミノ酸246-270、TRAF6は、230-245であった。またN末端側を欠失したTRAF5の発現はCD40刺激に依存したCD23の発現誘導をdominant negativeに抑制し、N末端側を欠失したTRAF6の発現は40刺激に依存したNFkB活性化をdominant negativeに抑制した。TRAF5及びTRAF6はCD40細胞質領域の独立した二つのNFkB活性化部位からのシグナルを各々伝達することが示唆される。
2.Bcl-3蛋白質による癌化機構:Bcl-3がLyn,Fyn,c-Srcと相互作用しそのキナーゼ活性を上昇させることを示唆する結果を得た。
3.アフリカツメガエルの胚発生におけるrel関連遺伝子産物の役割:
Xlyt10をプローブにwhole mount in situ hybridizationを行った。原腸胚では局在が見られなかったが、神経胚では壁側中胚葉に局在していた。この結果はRel関連蛋白質の分化における新たな機能を示唆している。
4.B細胞特異抗原CD40からのシグナルによるRelの活性化及びそのB細胞アポトーシスにおける意義:
CD40細胞質領域アミノ酸220-245及び246-270の二つの部位が独立にNFkB活性化シグナルを伝達することを明らかにした。
