細胞基質間接着制御機構の解明とその癌的増殖性への関与

研究概要

癌細胞の悪性増殖へ深く関わる細胞基質間接着制御機構及びそのシグナル伝達に関し、以下の諸点を明らかにした。
1。細胞基質間接着に伴って,tansin,Fak,paxillin等のインテグリン裏打ち蛋白質群のチロシンリン酸化が誘起されるが、この反応の制御には、リン酸化酵素よりも、脱リン酸化酵素が主体であることを示す結果を得た。この脱リン酸化酵素の見かけ上の活性は細胞基質間接着及び接着に伴うアクチンストレスファイバーのintegrityに制御されていると考えられる。
2。ヒトpaxillin isoformを2種単離し、これらが或種の癌細胞や単球細胞に発現していることを明らかにした。これらの新規isoformは単独で発現しているのではなく、同一細胞に複数のisoformが発現しており、各々異なった機能を持つことが明らかになった。
3。g isoformは細胞運動性に関係していることを示唆する結果を得ている。
4。paxillin isoformの更なる機能解析を行う為、マウスの遺伝子座位のcloningを行い、その構造解析を終了した。g isoformの遺伝子座位に関しては、マウスとヒトとで大きく異なる。
5。サイトカイン刺激がインテグリン裏打ち蛋白質群のチロシンリン酸化に作用し、細胞の形態・運動性を変化させることが知られているが、モデル系として、インシュリンを用い、インシュリン刺激によるインテグリン裏打ち蛋白質群のチロシンリン酸化制御にチロシンリン酸化酵素Cskが重要な役割を果たしていることを明らかにし、さらに、その分子機構を示した(東京大学医学部第三内科との共同研究)。
