研究概要	テロメラーゼは、細胞が無限分裂寿命をもつ(不死化する)ために不可欠の酵素と考えられ、正常体細胞組織にはなく、癌組織にはあることが予想されていた。本研究の目的は、1)テロメラーゼの精製とcDNAのクローニング、2)癌の発生・進展にかかわるテロメラーゼの癌組織および前癌病変に特異的な発現、3)テロメラーゼの発現調節を担う遺伝子の解明、4)テロメラーゼをターゲットとする癌の新しい早期診断法の確立、および5)新しい化学療法剤ターゲットとしてスクリーニング法の確立、である。本研究の成果として、以下のころが本年度に明らかになった。 1)テロメラーゼ活性は、癌組織では高頻度に強陽性であり、正常組織ではきわめて弱いか陰性であることが、多くの検体で確認された。2)初期の癌でもテロメラーゼ活性陽性頻度が高い場合が多いことがわかった。3)以上の知見は、テロメラーゼ活性が新しい癌診断の遺伝子マーカーとして期待できることを示した。4)頻度は低いが前癌組織でも高い活性を示すものがあった。5)癌診断および制癌阻害剤スクリーニングのターゲットとしてのテロメラーゼ活性測定のため、テロメラーゼ測定の簡易化、定量化などの改良が進められた。6)癌化の過程でのテロメア短縮とテロメラーゼ発現が、異常染色体をもった細胞を安定に存続せしめ、細胞癌化に寄与することが示唆された。7)テロメラーゼ活性を抑制する遺伝子がヒト3番および5番染色体上にあることがわかり、前者の位置は数Mbまでにせばめられた。8)ヒトテロメア結合蛋白質のcDNA候補を複数クローニングすることに成功した。7)癌細胞の分化によりテロメラーゼ活性が抑制されることがわかった。8)テロメラーゼ酵素の精製、cDNAクローニングにはまだ成功しなかった。
