癌細胞の増殖と転移を支える細胞外基質/インテグリン系の解析とその応用

研究概要

種々の細胞外基質と多様なインテグリンの相互作用は、細胞の接着固定、増殖、移動、分化等に重要な役割を果たしている。癌の浸潤・転移の過程において、癌細胞は自らの増殖や移動に都合のよい細胞外基質を利用すると考えられている。本研究では高い細胞運動促進活性をもつラミニン5(ラドシン)とその細胞表面受容体であるインテグリンα3β1に注目しながら、7名の班員がそれぞれ独自の立場から、癌細胞の増殖、浸潤、転移に関与すると思われる細胞外基質/インテグリン系分子の種類や作用を解析した。1)ラミニン5(LN5)のα3鎖のG領域には3個の細胞接着ドメインが存在し、G2はインテグリンα3を認識し、G4とG5は糖鎖を認識することが示された。一方、LN5の強制発現実験によって、LN5が腫瘍形成を促進することが示唆された。2)リポソーム化シアリルルイス-x(SLeX)が癌細胞の肺への集積を強く抑制し、一方、合成RGDS誘導体は標的臓器微小管の内皮細胞への着床後の癌細胞の基底膜浸潤を顕著に阻害することが明らかになった。3)インテグリンα5β1とフィブロネクチンとの結合親和性が、フィブロネクチンのEDA領域の選択的スプライシングにより制御されていることを明らかにした。4)ラミニン及びIGFI受容体を介したシグナル伝達系が平滑筋細胞の分化形質を維持することを明らかにした。5)マウスα3インテグリン遺伝子の転写調節領域を明らかにし、また形質転換細胞での転写亢進を確認した。6)インテグリン遺伝子の導入実験において、インテグリン分子の特性により臓器特異的な転移、転移性の亢進、転移性の抑制などが見られた。7)HB-EGF-DRAP27/CD9-インテグリン複合体に、TM4ファミリーに属するCD63とCD81および21kDの新規蛋白がアソシエ-トしていることを明らかにした。
