研究概要	アポトーシスはプログラム細胞死の一型で、生体の恒常性維持に重要な役割を果たしている。したがってアポトーシスの異常はがんを含めた様々な疾患の発生原因となる。一方細胞のがん化やがん細胞の転移のプロセスがアポトーシス耐性と密接な関係にあることもわかってきた。このようなことからアポトーシスに関する研究はがんの発生や進展・転移の機構を理解していく上からも非常に重要であると認識されるようになってきた。ところがアポトーシスに関する研究はこれまでFasやTNFRを介するものを中心に進められ、がん遺伝子やがん抑制遺伝子、抗癌剤など、TNFRファミリー蛋白質以外の因子によって誘導されるアポトーシスについてはほとんどまとまった情報がなかった。そこで本研究班では各班員が独自に開発した実験系を用いて、c-Mycやs-Myc,LPS,アデノウイルスE1A、p53の標的遺伝子であるbaxなどによって誘導されるアポトーシスにおけるシグナル伝達機構の解明を試みた。その結果TNFRファミリー蛋白質以外の因子によって誘導されるアポトーシスにおいてもFasやTNFRの場合と同じく、カスパーゼ3様プロテアーゼを主体とするプロテアーゼカスケードの活性化が要求されるということがわかった。またえられた研究成果をもとにがん細胞が獲得しているアポトーシス耐性の機構についても解析を行ったその結果例えばc-Mycと協調してT細胞性リンパ腫の発生に寄与しているPim-1には、これまでの予想とは異なり、それ自身のもつキナーゼ活性に依存してc-Mycからのアポトーシスシグナルを増強する能力のあることを突き止めた。また悪性リンパ腫細胞がリンパ球ストローマ細胞依存的に増殖できるのは、ストローマ細胞から分泌されるシステインやIL-7によって腫瘍細胞でのBcl-2の発現が亢進され、それによって細胞死が抑制されるためであろうということを明らかにした。このように本研究班はプログラム細胞死の全体像を分子レベルで理解し、がん細胞のもつ弱点を暴きそれを標的としたがんの治療法の開発を行っていくための研究に精力的に取り組んでいる。
