マウス発生工学を用いた核内レセプターRXRの機能解析

研究概要	レチノイン酸レセプターは核内レセプターファミリーに属する蛋白質であり、レチノイン酸レセプター(RAR)と、レチノイドXレセプター(RXR)の2群に分けることができる。このうち、RXRはホモ2量体を形成するほか、さまざまな核内レセプターと安定なヘテロ2量体を形成して機能することが知られている。in vitroではこのRXRのリガンドである9-cis retinoic acidがT細胞のアポトーシスを強く抑制することが報告されている。胸腺でのT細胞の選択、分化における核内レセプターの関与を検討するために、RXRβのdominant negative型を胸腺細胞で強く発現するトランスジェニックマウスを作製し、抗原特異的刺激を与えることにより検討を加えた。RXRβ cDNAのC端を欠失したdominant negative型(RXRβΔC2)をコードするcDNAを胸腺特異的なlckプロモーターの下流に結合した融合遺伝子を発現するトランスジェニックマウス(lck-RXRβΔC2)を作製した。このマウスとI-A拘束性にOvalbumin(OVA)によって胸腺内のT細胞の負の選択が誘導されるOVA-TCRトランスジェニックマウスとの交配を行い、ダブルトランスジェニックマウスを作製した。その結果、抗原特異的刺激による負の選択が、RXRのdominant negative型を胸腺細胞で発現するトランスジェニックマウスで増強されることを確認した。核内レセプターNur77は胸腺におけるT細胞の負の選択を促進すると考えられているが、RXRのdominant negative型を胸腺細胞で発現するトランスジェニックマウスで、このNur77の発現の増強がみられることから、RXRβは胸腺においてNur77と相互に影響しながらT細胞の負の選択によるアポトーシスを抑制している可能性が示唆された。今後、アポトーシスの経路のどの部分に関与しているのかを詳細に解析する予定である。
