温度感受性変異p53遺伝子を持つマウス赤白血病細胞の生物特性

研究概要	フレンドウイルスgp55遺伝子導入トランスジェニックマウス由来の赤白血病細胞系より温度感受性変異p53(p53^<val-135>)遺伝子を発現する赤白血病細胞株を樹立し、その亜株(El-gp55-1-2-3)を分離した。同細胞株では変異p53は細胞質に局在するが、温度シフトにより野生型p53の発現を誘導するとp53は核内に移行し標的DNAに結合する。即ち、1-2-3細胞を37℃、あるいは32℃で培養した後、抗p53モノクローナル抗体(PAb421)を用いて免疫染色を行った。37℃では細胞質が、32℃では核が染色された。さらに、野生型p53の発現に伴いこの赤白血病細胞系の細胞増殖能が顕著に低下し、アポトーシスが誘導されることを明らかにした。即ち、32℃で培養した場合アポトーシス特徴的な染色体DNAの断片化が観察された。またこの時、細胞周期制御に関与するp21、gadd45、およびcyclin Gの発現は活性化されるが、アポトーシス関連遺伝子bcl-2、bax、fas、およびfas ligandの発現上昇は認められないことを再現性よく確認した。p53遺伝子変異により細胞周期制御に関与する遺伝子の機能が抑制され、細胞の腫瘍性が維持される可能性が示唆された。この細胞系の腫瘍増殖シグナルはgp55が結合するEPORの下流にあるが、検索した結果JAK1,STAT5が恒常的にチロシン燐酸化していることが明かになった。しかしSTAT5は核内移行ができず、他の経路のシグナルを用いている可能性が示唆された。
