研究概要	ショウジョウバエMAPキナーゼキナーゼの優性活性化型変異によって表現形の回復されるP因子挿入突然変異Gp99を同定し、これがM期の進行に必要な新規の細胞周期因子をコードすることを、これまでに明らかにしてきた。データベースの検索から、Gp99のコードするタンパク質(462アミノ酸残基)の中央部の約150残基の領域に類似した配列を共通に持つ4種のタンパク質を、全ゲノム配列の決定された出芽酵母から見い出した。また、分裂酵母・線虫からもそれぞれ2種ずつ、ヒトからも1種の断片配列を見い出した。いずれも、塩基配列から推定された機能未知の予想タンパク質であり、ファミリーを構成することが明らかになった。本年度は、出芽酵母で見い出された、4種の相同遺伝子をクローニングし、その内の一つSc-1について、遺伝子破壊実験を行なった。その結果、生存に必須の遺伝子であり、G1期で停止することが明らかになった。このことから、酵母Sc-1もGp99と同様に細胞周期制御に関わることが示された。その作用する時期の違いは、Cdc2(出芽酵母ではCDC28)が、高等植物ではM期に作用するのに対して、酵母ではG1期とG2/M期の両期で作用するように、Sc-1もG1期と共にG2/M期にも作用することが考えられる。あるいは、各ファミリーメンバーがそれぞれ細胞周期の異なったポイントで作用することも考えられる。現在、他の遺伝子についても遺伝子破壊実験が進行中である。Two hybrid法を用いて、Gp99に結合する因子をコードするショウジョウバエcDNAのスクリーニングを行なったが、得ることができなかった。また、Gp99に対する抗体を作製し、分割期胚における分布を解析し、スピンドルに分布するという予備的結果を得た。
