c-kitレセプターチロシンキナーゼの恒常的活性化と腫瘍原性に関する研究

研究概要	c-kitレセプターチロシンチナーゼ(KIT)は、リガンド(stem cell factor:SCF)依存性に活性化される。このKIT/SCFシステムは、造血幹細胞、マスト細胞、生殖細胞や色素細胞の増殖・分化に必須である。我々は、c-kit遺伝子にリガンドが存在しなくても恒常的に活性化するgain-of function変異が存在することを見いだした。特に、kinase領域のAsp^<816>コドン(マウスではコドン814)は恒常的活性化変異のhot spotであり造血器腫瘍の発症に関与していると考えられる。一般に、KITは二量体化し細胞内にシグナルを伝達することが知られているが、kinase領域内のc-kit変異では細胞外領域におけるダイマー形成は認められなかった。kinase領域に変異によるKITの恒常的活性化や腫瘍化シグナル伝達系における細胞外領域の役割を検討するため、野生型(wild type)KIT^<WT>と恒常的活性化KIT^<Tyr814>の細胞外領域を欠失させたDel(Δ)KIT^<WT>とΔKIT^<Tyr814>を作製し、KIT陰性であるIL-^3依存性細胞株BA/F^3へ導入した。ΔKIT^<WT>導入BA/F^3細胞では、ΔKIT^<WT>のチロシンリン酸化や活性化は認められず、IL-^3非存在下では増殖反応を示さなかった。一方、ΔKIT^<Tyr814>導入BA/F^3細胞では、ΔKIT^<Tyr814>の恒常的活性化が認められ、導入細胞もIL-^3非依存性に増殖した。このΔKIT^<Tyr814>によるIL-^3非依存性増殖は、KIT^<W42>(KITのdominant negative型)によりほぼ完全に抑制された。さらに、ΔKIT^<Tyr814>は、KIT^<WT>やKIT^<W42>と共沈することが明らかとなった。以上の結果より、kinase領域のc-kit変異によるKITの活性化やシグナル伝達系には細胞外領域は必須ではないが、モノマーで作用するものではなく細胞内領域における相互作用が重要であると考えられた。
