細胞内増殖機構における蛋白質リン酸化酵素PKNの機能に関する研究

研究概要	本研究においては、PKNと直接的、間接的に相互作用することによりシグナリングを制御している分子の同定を種々の方法を用いて行い、既知のシグナリング分子との関連も含めて、複雑なシグナリングネットワークの解明の一助とすることを目指した。以下に、得られた結果の概略を記す。 1.PKNと低分子量GTP結合蛋白質Rhoとの相互作用に関する解析細胞骨格の構築に重要な役割を担っていると考えられる低分子量GTP結合蛋白質PhoがPKNと直接GTP依存的に結合し、PKNの活性化を引き起こすことを示し、Rhoの標的蛋白質の一つとしてPKNが機能している可能性を示した。PKNとRhoの結合について、酵母Two hybrid systemなどを用いて詳細に検討した結果、Rhoとの結合にはPKNのアミノ末端側領域中に存在するロイシンジッパー様配列が重要であることを明かにした。 2.PKNと相互作用する蛋白質の解析 PKNと相互作用する蛋白質を酵母Two hybrid systemによりスクリーニングすることにより中間径フィラメントであるニューロフィラメントとPKNは直接結合し、またこれをリン酸化することによりニューロフィラメントの結合、脱重合を制御していることを示した。さらに他の中間径フィラメントであるビメンチンともPKNは結合することから、PKNはこれら中間径フィラメントのダイナミックな動きを制御している可能性が示唆された。また、上記と同様な方法で、細胞骨格蛋白質アクチンの重合に重要な機能を持っているα-アクチニンともPKNが直接結合することを示し、その結合領域、結合様式などを詳細に検討した。 3.PKNの細胞内局在に関する解析 PKNに対する特異抗体を用いてPKNの細胞内局在について解析したところ、通常の状態ではPKNは細胞質中に存在するのに対して、熱などのストレスにより速やかに核に移行することが明かとなり、PKNは、遺伝子発現制御を含む核内でのイベントにも何らかの機能を持っている事が示唆された。
