研究概要	サイトカインはホルモン様生理活性物質であり、サイトカインの産生異常により様々な炎症疾患、免疫、血液異常が起こる。正常細胞の癌化、癌細胞の増殖・転移に伴う悪液質、免疫不全症にもサイトカイン産生異常が深く関与するものと推定されている。本研究班は1)癌増殖、転移、病態調節に関わるサイトカインネットワークの解析2)サイトカイン産生制御機構、受容体シグナル伝達機構の解析3)サイトカイトを分子標的とした癌免疫治療に関する基礎研究を目的とした。今年度の主な研究成果は以下のように要約される。1)マウス悪液質誘導性大腸癌株による悪液質がIL 10遺伝子導入により抑制できることを明らかにした。さらに、悪液質誘導性株に特異的に表現する遺伝子として新規のメタロプロテアーゼ(ADAM-TS1)遺伝子をクローニングした。2)IL 12投与担癌マウス脾細胞は強い腫瘍浸潤能を有すること、IL 12投与によりマウス腫瘍部位における血管内皮細胞のICAM-1,VCAM-1発現が亢進することを明らかにした。3)STAT3遺伝子欠損マウスを作製することにより、STAT-3が初期発生に必須の分子であること、IL 6のSTAT-3活性化にはSTAT-3のC-末端部位が関与することを明らかにした。4)NF-IL 6,NF-IL 6betaダブルノックアウトマウスで脂肪細胞の分化が障害されることを明らかにした。
