プロテインホスファターゼの特異的阻害蛋白遺伝子によるがんの細胞工学的制御

研究概要	細胞の癌化はリン酸化酵素の異常のみならず、脱リン酸化酵素の異常によってもおこることが近年明らかにされた。我々はラットの低分化型腹水肝癌細胞でプロテイチンホスファターゼPP1が著しく高発現していることを見いだした。そこでPP1活性を抑制することががん治療につながる可能性が考えられた。本研究ではプロテインホスファターゼPP1の特異的阻害蛋白Inhibitor-2(1-2)の遺伝子導入により癌細胞の増殖を制御することを目的とし、以下の成果を得た。 (1)ヒトB細胞白血病細胞株Daudiから1-2のcDNAをクローニングし、大腸菌で発現させて精製し、それでウサギを免疫し特異抗体を得て以下の実験に用いた。 (2)1-2cDNAを導入したCOS-7細胞はコントロールの5-10倍の1-2を発現し、その抽出液のPP1阻害活性はコントロールの2倍以上に増加し、PP1活性は40%減少していることから、導入・発現した1-2が細胞内で機能していることが明らかとなった。 (3)ヒト1-2の発現ベクターをマウスBリンパ腫由来細胞WEHI-231に導入し1-2を発現させた。この時ネオマイシン耐性クローンのうちで1-2を発現するクローンの割合は2/70と極めて低く、その陽性の2クローンにおいても導入DNAによる1-2の産生量は少ないことから、1-2の過剰発現は致死的であることが示唆された。さらにこの1-2発現株は増殖が遅いことからも1-2発現によりWEHI231の増殖が抑制されたと考えられる。今後は種々の細胞株を用いて、この1-2導入による増殖抑制効果の細胞特異性を明らかにすると共に変異性1-2発現ベクター等を用いて癌細胞に対する選択性の高い増殖抑制法を開発する。
