肺がんの薬剤耐性と転移因子を分子標的とした免疫遺伝子治療

研究概要	臨床を反映した肺癌の遠隔転移モデルの作製と癌関連抗原を標的とした分子生物学的制御を目的に検討し、以下の諸点を明らかにした。 1.NK活性欠損のSCIDマウスに組織型が異なるヒト肺癌細胞を経静脈的に移入し、6-8週間後に多臓器への転移形成モデルを確立した。また、左心室腔内への接種による骨転移モデルを確立した。 2.M-CSF、IL-10,MCP-1遺伝子導入ヒト肺癌細胞の転移能を上記モデルで検討しMCP-1遺伝子導入肺癌細胞は何ら転移形成に影響しなかったが、M-CSFおよびIL-10遺伝子導入肺癌細胞は転移臓器の微小環境条件により異なった転移形成能を示した。 3.抗P糖蛋白抗体はNK細胞、マクロファージの抗体依存性細胞傷害反応(ADCC)を誘導し、NK欠損SCIDマウスでの多剤耐性ヒト肺癌転移モデルにて治療効果があることを示し、さらに癌細胞へのMCP-1またはM-CSF遺伝子導入が抗P糖蛋白抗体の治療効果を増強することを明らかにした。多剤耐性癌細胞へのIL-2遺伝子導入もヌードマウスへの皮下移植モデルで抗体治療を増強した。 4.ヒト小細胞肺癌が高発現しているガングリオシドGM2に対するマウス・ヒトキメラ抗体KM966がADCCを誘導しヒト小細胞性肺癌細胞の遠隔転移形成を抑制し、生存期間を延長した。 5.肺癌浸潤による胸膜炎のIFN-γ局所療法効果、キラー誘導サイトカイン(IL-12,IL-15)による肺癌患者リンパ球キラー活性発現とTh2サイトカイン産生能を示した。以上、肺癌転移制御のため癌関連抗原に対する抗体を中心に、サイトカインおよびサイトカイン遺伝子導入の併用による集学的治療の可能性を検討してきた。今後は、骨転移、脳転移モデルでの検討、さらにin vivoでのサイトカイン遺伝子のキャリヤ-として効率のよいベクターの開発を進めていく予定である。
