ヒト唾液腺の癌細胞におけるベスナリノンの抗腫瘍活性

研究概要

ヒト唾液腺癌細胞(TYS)を、新規の細胞分化・アポトーシス誘導剤であるベスナリノン(3,4-dihydro-6-[4-(3,4-dimethoxybenzoyl)-1-piperazinyl]-2(1H)-quinolinone)で処理して認められる抗腫瘍効果を解析し、以下の諸点を明らかにした。
(1)TYS細胞のp53は、コドン281^<Aep-Hs>で点変異の生じていることをPCR-SSCPとdirect DNA sequencingにより明らかにした。
(2)TYS細胞をベスナリノンで処理すると、腺房細胞への分化とアポトーシスが生じ著明な抗腫瘍活性が認められ、それに伴って、p21^<WAFI>とTGF-β1との発現がup-regulationされることをNorthem blottingとimmunoblottingにより検出した。
(3)TYS細胞において、ベスナリノンによるp21^<WAF1>の発現誘導は、シクロヘキシミドで阻害されないことより、ベスナリノンは直接的にP21^<WAF1>を誘導していることが示唆された。また、TGF-β1でTYS細胞を処理するとp21^<WAF1>の発現誘導が認められた。
(4)TYS細胞のベスナリノン処理により、培地へのlatent TGF-β1の産生の増強がELISAにより検出された。
(5)TYS細胞において、TGF-βレセプター(I.II,III)の発現がaffinity cross-linkingにより検出された。
(6)ベスナリノンで処理したTYS細胞において、増殖抑制に伴い発現誘導される遺伝子として、TSC-22遺伝子のヒトホモローグのcDNA断片を得た。更に、そのcDNAの完全長をクローニングし全塩基配列を決定した。すなわち、ベスナリノン処理TYS細胞より、anti-sense oriented cDNA libraryを構築し、このcDNA libraryにより形質転換された大腸菌XLOLR株のコロニーをランダムに選択し、アルカリ法によりphagemidを調製した。更に、ジデオキシ法によりcDNA断片の一部の塩基配列を決定し、ホモロジー検索を行った。ランダムシークエンスで得られたヒトTSC-22 cDNAは1313bpでopen reading frame(ORF)を完全には含んでいなかった。そこで、phagemid libraryを用いて5'-RACE変法によりヒトTSC-22完全長cDNAをクローニングした。
