HLA-A31拘束性胃癌拒絶抗原ペプチドの癌ワクチンとしての有効性に関する研究

研究概要	CD8+キラーT細胞より認識されたHLA-A31により提示されるヒト胃癌拒絶抗原のnatural antigenic peptideと思われる分子を単離した。 1.一次構造はTys-Ser-Trp-Met-Aap-Ile-Ser-Cys-Trp-Ileの10ヶのアミノ酸よりなり(F4.2と命名)、N末3番目、6番目はA31結合モチーフと合致したアミノ酸であり、これらがアグレトープとしてA31に結合する主たるアミノ酸と推測された。 2.C末より2ヶアミノ酸を除くとキラーT細胞は反応せず、従って、N末より9番目のTrpがT cell epitopeと推測された。 3.この抗原は他の胃癌(MKN28)にもHLA-A31を発現させると拒絶抗原として働いた。しかも、この構造はF4.2と類似したものであることが推測された。従って胃癌ワクチンとしての可能性が考えられた。
