ヒト胃の酸・ペプシ-ノーゲン分泌細胞の水チャネルのクローニングおよび機能の解析

研究概要

筆者はヒト胃で特異的に発現している水チャンネルを発見した。これは、ラット脳において発現が確認されているAQP4と近縁の水チャンネルで、興味深いことに、消化管の中では胃にのみ発現しており、小腸や大腸では全く発現が確認されなかった。さらに胃を構成する細胞の中でも酸分泌細胞(壁細胞)およびペプシノーゲン分泌細胞(主細胞)に局在し、他の上皮細胞や粘液分泌細胞には存在しなかった。これらのデータより、AQP4は食物の初期消化のための胃液分泌に深く関与しているであろうことが示唆された。また最近、ラット胃においてもAQP4の存在が確認され、その発現部位も壁細胞に特異的であったことより、AQP4の消化への関与は哺乳類に広く保存されている形質であろうと推定した。次にAQP4の生理学的、生化学的実験を個体レベルで詳細に行うために、これと相同なラット胃の水チャンネルcDNAを得、in vitro transcripitionによってcDNAを作製し、アフリカツメガエル卵母細胞へ打ち込み、細胞内外の浸透圧差に依存した水の透過性を測定した。さらに構造・機能相関について調べるため、delection、あるいはsite-directed mutationによりどの部分がチャンネルとしての機能に重要であるかを明らかにし、特にリン酸化によるチャンネルの水透過性や細胞内局在の変化についても検討を行い、リン酸化により細胞表層膜に移動することが明らかになった。
