植物液胞膜水チャネルとプロトンポンプの分子構造と機能調節

研究概要	まず水チャネルの分子構造をcDNAクローニングを通して明らかにした。高純度の液胞膜が多量に得られるダイコンを材料とし、γ-VM23とδ-VM23と名付けた分子のcDNAを得て解析した。2つのmRNAをツメガエル卵に注入し、水チャネル機能を測定した(共同研究:生理学研究所岡田泰伸教授、森島繁博士)。γ-VM23のmRNA注入した卵では83.9x10^<-4>cm/s、δ-VM23は67.1x10^<-4>cm/sであり、水を注入した卵に比べ7倍以上の水透過率の上昇が見られた。この結果は2つが水チャネルであることを示している。γとδのアミノ酸配列は60%が一致した。γ-VM23は根、茎、葉いずれでも発現していたが、γ-VM23のmRNAは根には検出できなかった。植物の地上部と地下部で水チャネルの発現調節が異なるものと推定される。ダイコンの細胞質膜型水チャネルの構造も明らかにした。細胞質膜型水チャネル(PAQ1)はアミノ酸286個からなり、液胞型よりも30数残基N端側部分が長い。液胞型分子と比較すると、γ-VM23とは36%、δ-VM23とは38%のアミノ酸が一致するのみであった。今後細胞質膜と液胞膜の各分子を比較しつつ、各分子の協関、機能分担を明確にしたい。さらに、液胞膜Ca^<2+>-ATPaseとCa^<2+>/H^+対向輸送体に関する解析を行った。植物細胞の主要な細胞内Ca^<2+>プールは液胞である。液胞へCa^<2+>を能動輸送する装置として、Ca^<2+>-ATPaseとCa^<2+>/H^+対向輸送体の酵素学的性質を明らかにした。液胞膜Ca^<2+>-ATPaseはATPのみを基質とし低Ca^<2+>条件で高い活性を発揮する。一方、Ca^<2+>/H^+対向輸送体は10μM以上で高い活性を示す。生理的な細胞質Ca^<2+>濃度を考慮すると、Ca^<2+>-ATPaseが主要なCa^<2+>輸送体として機能し、対向輸送体は細胞質Ca2+を1μM以下に維持する役割を担っていると考えられる。
