バクテリオロドプシンのプロトントランスポータとチャネル

研究概要

本研究の目的は光エネルギーを使ってプロトンを1方向に運ぶ膜蛋白質、バクテリオロドプシンのポンプメカニズムを、シッフ塩基部分(トランスポータ)とプロトン輸送路(チャネル)との相関に焦点を絞って明らかにすることである。特に我々のグループが開発した、赤外分光法により蛋白質に結合した水分子を直接観測する手法を駆使して、トランスポータがどのようにチャネルを制御するかを検討してきた。本年度は以下のように新たな水分子の局在が明らかになり、バクテリオロドプシンにおけるトランスポータとチャネルとの機能的相関に対する理解を深めることができた。
(1)プロトンではなくクロライドを輸送するバクテリオロドプシン変異体(D85T)の活性中心では、クロライドが水分子に水和された構造をとることがわかった。活性中心における負電荷の水和はバクテリオロドプシンにもみられ(この場合Asp85)、プロトンポンプとクロライドポンプに共通の構造を示唆する。一方、クロライドポンプの変異体ではシッフ塩基の水素結合の弱いことがわかった。
(2)細胞外側ドメインに存在するArg82,Glu204の変異体を用いた研究から、この領域に存在する水分子が同定された。プロトン放出基であるGlu204からのプロトンの解離を制御するArg82は、プロトンポンプの前後でトランスポータ部位との相互作用を変化させること、このとき水素結合構造が重要な役割をはたすことが明らかとなった。
(3)細胞質側ドメインに存在するVal49の変異体を用いた実験から、トランスポータと細胞質側のAsp96を結ぶ経路には、Val49のペプチドC=Oを仲立ちとした水素結合ネットワークが存在することが明らかになった。この水素結合には水分子も関与しており、このネットワークが遠隔的な相互作用によるプロトン結合基(Asp96)のpKa変化をもたらすものと考えられる。
