研究概要	本分担研究では、イオンポンプのうちサブユニット組成が単純なNa/K‐及びCa‐ATPaseに着目し、これらイオンポンプ間の“キメラATPase"を用いた分子生物学的、電気生理学的手法、及び原始間力顕微鏡(AFM)を用いた形態学的手法使って、機能ドメイン(特にイオン輸送ドメイン)の同定を目指す。これまでに以下のことがらを明らかにした:(1)Ca‐ATPaseには、Ca‐感受性部位以外に、Na‐及びK‐感受性部位が存在し、ATPase活性、イオン輸送活性に重要な役割を果たしている(雑誌論文1、図書2)。(2)Na/K‐ATPaseの疎水性領域とCa‐ATPaseの親水性領域とから構成されたキメラはK‐イオンを内向き輸送する。従って、疎水性膜貫通領域には複数のイオン結合部位が存在し、これらが共同してイオン輸送に関与するものと考えられる(原稿準備中)。(4)新しいP‐型ATPase4種を含む計12種のP‐型ATPase遺伝子をテトラヒメナからクローニングした(雑誌論文2)。(5)ニワトリとショウジョウバエのNa/K‐ATPaseにおいてもFoF1ATPaseのFo(H‐ポンプ)においてもpore構造が存在することを示し、Foのサブユニット構造も提唱した(雑誌論文3)。(7)DNA‐タンパク質複合体を用いてAFMの定量的解析に於ける応用性を確立した。タンパク質の大きさの決定、重合度の検討、特異的高次構造の検出等にAFMは有効であることが明らかになった(雑誌論文4と5)。
