AIDS粘膜ワクチン:Th1/Th2型細胞によるHIV特異的粘膜免疫の誘導

研究概要	AIDS予防対策を考えたとき、現在最も問題になるのが性行為を介したHIV感染である。HIVの最初の侵入が上皮細胞層でおき、そして全身系の免疫システムの破壊につながっていく。AIDSワクチン開発を進めるにあたってはHIVのこれらの感染経路を考慮した粘膜系と全身系両者の免疫機構を使った二段構えの防御システムを宿主側に成立させる必要がある。その目標に向かって本研究班は、粘膜免疫を応用したAIDS粘膜ワクチン開発のための基礎的新情報の収集を推進している。マウスを使用して、HIV/SIVを含めたタンパク質抗原やペプチドを粘膜免疫調節因子/粘膜アジュバントであるcholera roxin(CT)と併用して経口免疫すると抗原に特異的なIgG1とIgAが血清中と分泌液中に誘導される。さらに抗原特異的CD4^+T細胞のサイトカイン産生パターンを解析すると、IL-4、IL-5、IL-6に代表されるTh2型サイトカインが顕著に誘導されていることが解った。つまり、経口免疫(抗原プラスCT)により誘導されたTh2型CD4^+T細胞がIgG1とIgAを誘導している。これらの実験結果を基に、ヒトにより近い霊長類のモデルとして、SIV感染モデルに使われるアカゲザルをSIVp55抗原とCTの混合ワクチンで経口免役し、抗原特異的Th1/Th2型と抗体免疫応答を細胞・分子レベルで解析した。経口免疫後、SIV特異的IgGとIgAが血清中と分泌液中に誘導され、二重の免疫が成立していることが示唆されている。さらに抗原特異的CD4^+Th細胞の解析では、分泌型IgAを誘導するために必要なTh2型細胞だけではなく、HIV感染阻止を考えたときに必要な細胞性免疫の誘導を示唆するIFN-γ産生Th1型細胞も誘導されていた。HIVの侵入門戸である粘膜面に抗原特異的な細胞性免疫と液性(分泌型IgA)免疫の両方が経口免疫により誘導出来る可能性が強く示唆された。
