研究概要	Tリンパ球細胞に潜伏感染しているHIVは、リンフォカイン産生を誘導するような物質(PHAやTPA)により活性化され、ウイルス産生、AIDS発症に至る。現在この現象の機構としてはTPAなどによりHIVのエンハンサー結合蛋白質など宿主細胞側の転写因子が活性化されHIVプロウイルスの遺伝子発現が誘導されるためと考えられている。本研究ではHIVのLTR(long terminal repeat)領域に結合するMybの宿主細胞側の転写因子の構造・機能を解析し、その活性制御機構を明らかにし、HIVプロウイルスの遺伝子発現誘導を阻止する薬剤開発の基礎とすることを目的とした。myb遺伝子産物(Myb)はHIVやLTR領域内の3ヵ所に結合し、HIVの遺伝子発現を促進する。しかし、どのようなメカニズムでMybが転写を活性化するかについてはこれまで全て分かっていなかった。Mybなどのエンハンサー結合蛋白質が転写を活性化するためにはTBP/TFIIB/RNAポリメラーゼなどの基本転写因子と相互作用する必要がある。しかしMybなどの因子と基本転写因子とが直接結合することはなく、両者の間にはコアクティベータ-と呼ばれる因子が存在し、ブリッジの役割を果たすことが、基本転写因子の研究から分かりつつある。そこでMybによる転写活性化メカニズムの解明にはMybのコアクティベータ-を同定することが重要となる。私達は転写因子MybがHIV LTRからの転写を活性化するためにはコアクティベータ-CBPが必要であることを明らかにした。CBP分子内の同じ領域に2つの転写因子CREBとMybが結合することから、両者のCBPへの結合が競合することによって、互いの活性を阻害し合う可能性が考えられる。実際に私達の最近の結果では、Mybの活性がCREBやcAMPの添加によって阻害されることが示されている。今後、細胞内cAMP濃度を上昇させる種々の薬剤がHIVの転写およびHIVプロウイルスの産生に及ぼす影響を調べることは有意義であると思われる。また、CBPの同定によって、Myb活性を阻害するための薬剤開発の新たな局面が開かれた。
