中枢神経機能におけるプロスタグランジンE受容体の役割

研究概要	EP3受容体サブタイプの神経細胞における新規の機能 4つのサブタイプの中で、EP3受容体は中枢において最も発現量が高くかつ幅広く分布しているサブタイプである。神経細胞におけるEP3受容体の機能を調べるため、PC12細胞にEP3受容体を発現させて細胞の形態日対する作用を調べた。EP3受容体は、PC12細胞でGiを介してアデニル酸シクラーゼの活性を抑制したが、細胞内Ca^<2+>の動員は起こさなかった。一方、EP3受容体を発現したPC12細胞をNGFで分化させた後、EP3アゴニストは神経突起の退縮と成長円錐の消失を引き起こした。この作用は、百日咳毒素(PT)では遮断されなかったので、GiやGoを介した作用ではない。一方、ボツリヌス毒素C3酵素処理により完全に阻害されることから、その作用は低分子量G蛋白質Rhoを介していることがわかった。更に、Cキナーゼを活性化するフォルボールエステル、TPAも同様にRhoを介して神経突起の退縮と成長円錐の消失を引き起こした。細胞をTPAで長時間前処理し、Cキナーゼをダウンレギュレーションさせると、TPAによる神経突起の退縮と成長円錐の消失は完全に阻害されたが、EP3受容体による作用は全く影響を受けなかった。これらのことから、Rhoを介した神経突起の退縮と成長円錐の消失には2つの経路があり、1つはCキナーゼを介する経路で、もう1つはCキナーゼを介さない経路である。EP3受容体はGiを介してアデニル酸シクラーゼの活性を抑制する一方で、PT非感受性のG蛋白質を介してCキナーゼ活性化とは異なる経路でRhoを活性化して、神経細胞の形態を制御している。現在、後者の全く新しい情報伝達系路に関与する三量体型G蛋白は不明であるが、最近G12、G13がRho活性化するという報告があり、これらのG蛋白も含めてその同定を行っている。
