下オリーブ小脳投射系(登上線維-プルキンエ細胞間シナプス)再形成のメカニズム

研究概要	本研究では、単一軸索形態再構築法により、下オリーブ核ニューロンの軸索(登上線維)の側枝の発生とシナプス再形成の過程の特性とメカニズムを明らかにしようとした。実験には、正常ラットと、下オリーブ核に対する毒性物質である3-アセチルピリジン(3-AP)投与によって生き残った登上線維にシナプス形成を促したしたラット(3-APラット)を用いた。ビオチン化デキストランアミンの下オリーブ核注入により、下オリーブ核ニューロンの軸索を標識し、小脳・延髄の連続切片の組織標本から、顕微鏡下に、標識された単一軸索の形態を三次元的に再構築した。まず、正常ラットでは1個のニューロンの軸索あたり3から18本の登上線維が、ほとんどが白質で、分岐することが分かった。1本の枝は1個のプルキンエ細胞のみに投射していた。また、顆粒層および小脳核に投射する細い側枝がすべての軸索で認められた。3-AP動物での単一軸索再構成の結果では、次の所見が得られた。(a)小脳皮質の顆粒層・小脳白質における軸索側枝の数は、正常動物と比較して明らかな違いは無かった。(b)1本の登上線維が分子層内で軸索側枝を延ばし、周辺の数個から十数個のプルキンエ細胞にシナプスを形成するようになるのが観察された。これはRossiらの結果を確認したことになった。(c)小脳核側枝においてもシナプス再形成があるようで、小脳核側枝1本あたりの終末の個数が3-AP動物では約300個で、正常動物の約100個と比較すると約3倍に増加していた。以上の本研究の結果は、3-AP動物における軸索側枝発生・シナプス再形成は、軸索の途中から新たな枝が出るのではなくて、主として軸索の終末部の範囲で起こるということを示唆した。
