分裂酵母rad21^+遺伝子産物の核構造維持と染色体分配に関する機能の解析

研究概要

分裂酵母のrad21^+遺伝子は、DNAの二重鎖切断修復と細胞増殖に必須な遺伝子である。rad21^+遺伝子の生育に必須な機能を明らかにするため、我々はrad21温度感受性株(rad21-k1)を単離し、その解析を行った。その結果、rad21-k1株では、制限温度下で、短い微小管と凝縮した染色体をもつ細胞や、姉妹染色体が完全に分離せずに微小管によって引き伸ばされたり不均一に分配された細胞が出現し、さらにcut様の細胞や核が細胞の片側にdisplaceしたものが観察された。したがって、rad21^<1+>遺伝子産物は、特に分裂期の中期から後期において、染色体の正常な分配に必要であると考えられる。またこの変異株は、許容温度でUVやγ線に感受性を示すが、チューブリンの重合阻害剤であるthiabendazoleに対しても感受性を示し、またα-チューブリンの低温感受性変異nda2-KM52と合成致死になることなどから、rad21^+遺伝子産物は微小管に関わる機能も有することが分かった。さらにFACScanの解析から、rad21-K1株では、制限温度下でDNA複製は正常に起こっているが、許容温度においてDNA合成を阻害するhydroxyurea存在下、その生育は野生株に比べて阻害されたことから、DNA複製もしくはそのチェックポイントに何らかの関与があると思われる。興味深いことに、rad21-K1変異は、20Sサイクロソームを構成するCut9をコードするcut9^+遺伝子の変異とも合成致死性を示した。20Sサイクロソームはユビキチンリガーゼ活性をもち、サイクリン分解や分裂中期/後期遷移に必須である。rad21^+とcut9^+との遺伝的相互作用から、rad21^+遺伝子産物がユビキチン依存性蛋白質分解の経路を通じ、多様な細胞機能を果たしている可能性が考えられる。
