研究概要

複製開始領域の同定・単離とその特性ショウジョウバエの培養細胞を用いてDNAポリメラーゼα遺伝子近傍にA/Tに富む特異的な複製開始領域が存在することを、競合的PCR法を用いて明らかにした。また初期胚と培養細胞で複製開始領域を比較した結果、複製開始が転写活性と密接な関係にあることが認められた。出芽酵母を用いチェックポイント因子によるS期後期複製開始点の制御が、普遍的なものであることを第五染色体上の後期複製開始点を用いて明確にした。さらにチェックポイント機能が欠損している変異株では、S期後期に複製を開始する複製開始点からの複製開始が野性株と比べて早く起こることを明らかにした。複製開始蛋白質の同定とその作用機構分裂酵母Orcサブユニットであるorp1蛋白質が分裂酵母第2染色体上に同定した自立複製配列に特異的に結合していることを証明した。さらに、分裂酵母Mcm2と6蛋白質が、細胞周期特異的に核内の特定の染色体領域に結合していることを明らかにした。分裂酵母及び哺乳動物細胞より、Cdc7-Dbf4キナーゼ複合体の機能的ホモログを単離し、その機能解析を行った。その結果、両遺伝子とも細胞の増殖に必須であり、染色体複製に要求されることが明かとなった。アフリカツメガエル卵無細胞系を用いてS期特異的に起こるMcm2のリン酸化部位を同定した。複製開始に関わると考えられるツメガエルCdc45及びCdc14ホモログを同定し、Cdc45については複製開始に必須な因子であることを明らかにした。複製関連遺伝子群の発現制御機構DREFの異所的な発現をする遺伝子導入ハエを樹立し、DNA複製や細胞増殖の異常を詳細に解析した。その結果DREFがショウジョウバエのDNA複製開始において必須の機能を持ち、E2Fと協同的に働いて、S期への進行を制御していることを明らかにした。
