研究概要

真核生物染色体DNA複製反応には三つの機能の違ったDNAポリメラーゼα、δ、及びεが必須である。このうちで特にLagging鎖合成装置の中心的働きをしているDNAポリメラーゼδの複合体の構成が未だ明らかになっていない。遺伝学的手段が十分活性でき染色体DNAの全塩基配列が決定されている出芽酵母を使用してその複合体を精製して解析した。その結果、その複合体は主に三種類の蛋白よりなり、第一番目の125-KDa蛋白はCDC2遺伝子、第二番目の55-KDa蛋白はHYS52遺伝子、第3番目の48-KD蛋白はRFP3遺伝子でコードされていることが明らかに成った。一方、プライマー鋳型DNAを認識しそれに結合しDNAポリメラーゼδ及びεを助けてDNA鎖伸長を行うRF-C複合体の1つであるRFC2蛋白の温度感受性突然変異株を単離し、この蛋白がRfc5蛋白と同様染色体DNA複製のみならず細胞周期チェックポイント制御に関与していることを明らかにした。一方、ヒトの細胞でRF-C複製複合体と一緒に働いている複製因子PCNAの機能が、細胞周期制御因子p21蛋白によって制御される仕組みを明らかにするため、多数の変異型PCNAを作製しPCNA上のp21結合部位の解析を行った。染色体DNA複製には二本鎖DNAをunwindingするDNAヘリカーゼが必須であるが、その実体は未だ明らかではない。マウスの細胞からDNA複製の温度感受性変異株tsFT848細胞を分離し、解析することによりDNAヘリカーゼBが哺乳類細胞のDNA複製に関与するDNAヘリカーゼであることが明らかになった。この変異はDNAヘリカーゼBの724番目のアミノ酸であるPheからCysに変化していることを明らかにした。また、このヘリカーゼBの特異的抗体を作製して、DNAヘリカーゼBが複製の"場"に局在することが明らかになった。
