研究概要

遺伝学的及び生化学的手段を用い、Lagging鎖合成装置の中心的働きをしているDNAポリメラーゼδの複合体の構造は少なくとも三種類の蛋白よりなり、第一番目の125-KDa蛋白はCDC2遺伝子、第二番目の55-KDa蛋白はHYS2遺伝子、第三番目の48-KDa蛋白はPOL32遺伝子でコードされていることを明かにした。一方、Leading鎖合成装置の中心的働きをしていると考えられるDNA PolymeraseII(ε)複合体の最小サブユニットであるDpb4の機能を明らかにする目的で、この蛋白をコードするDPB4遺伝子を単離し、その破壊株を作成し表現型を調べた。その結果、破壊株は生存可能であり、ヒドロキシ尿素にも耐性を示すことが明らかとなった。同様にDNA PolymeraseII(ε)複合体と相互作用して、DNA複製に関与している蛋白をdpb11-1変異と合成致死となる変異のスクリーニングを行なうことにより新たにSld2、Sld3、Sld4(Cdc45)、Sld5、Psflなどを同定した。これらの蛋白の温度感受性突然変異dpb11-1、sld2-6、sld3-1、sld5-1、psf1-1株全てにおいてDNA複製開始及び鎖伸長反応が非許容温度下で著しく阻害されること、又複製開始に必要と考えられているCdc45とDpb11やSld3が相互作用していること、またCHIP assayからDNA PolymeraseII(ε)複合体がG1/S境界期から複製開始部位に結合し始めることなどからDNA PolymeraseII(ε)複合体はDNA複製開始の非常に早い段階からDNA鎖伸長反応に関与しているが明らかになった。Chromosome spread法により、各種のポリメラーゼやその他の複製蛋白の核内での局在解析を行ない、複製に関与しているポリメラーゼは、細胞周期を通じて核内で、核当たり20〜30個のFoci状に存在していることが明らかになった。。また、二重染色により、各々のポリメラーゼや他の複製関連蛋白との局在を比較したところ、S-期においてごく少数のポリメラーゼδとεのFociの一致が観察されたが、大部分のFociは別々に存在していた。複製ライセンシング因子であるマウスMCM蛋白質の遺伝的、生化学的解析を行い、ヒトMcm4,6,7複合体と同様、MCM4,6,7からなる6量体がDNAヘリカーゼ活性を内在的にもつことを証明した。一方、マウスDNAヘリカーゼBの核内での存在様式を詳細に解析することにより、DNAヘリカーゼBはDNA複製の開始か、開始に必要なクロマチン構造の変化に関与している可能性が示唆された。ヒトクランプ分子、PCNAとそのローディング因子、RFC間の複合体形成過程を、表面プラズモン共鳴、電気泳動、電子顕微鏡で解析した。どのようなリン酸化がRb蛋白によるDNAポリメラーゼα促進に重要であるか検討した。その結果、cdk2/cyclineEによってリン酸化されたRb蛋白がDNAポリメラーゼαの活性を最も促進した。又、DNAポリメラーゼαの4つのサブユニットの分子構築とドメイン構造を明らかにした。酵素活性に必要な最小領域は触媒サブユニットの330-1279アミノ酸であり、このドメインに隣接するC末端領域に他の3つのサブユニットが結合していた。
