研究概要	平成8年度より4年間行われる重点領域研究「細胞複製装置:染色体の複製・分配機構」の準備のために、計画研究の予定班員を中心にワークショップ「細胞複製装置:染色体の複製・分配機構」(1泊2日)を大阪大学医学部キャンパスにて開催した。ワークショップの内容は下記のとおりである。(1)真核生物の染色体複製開始領域の同定・単離とその特性の解析、(2)複製開始タンパク質の同定と作用機構、(3)In vitro DNA複製開始反応系の確立、(4)In vitro染色体複製装置の再構築、(5)複製関連遺伝子群の発現制御機構、(6)複製終結と姉妹染色体分離の分子機構、(7)染色体分配の分子機構、(8)染色体分配と細胞分裂の制御機構。各人が今までの研究成果の概要を発表し、さらに今後の研究計画について話し合った。研究項目A「染色体複製開始の制御機構」の研究分野では、染色体複製の開始に働く蛋白群が出芽酵母、分裂酵母、アフリカツメガエル、哺乳動物の細胞等で明らかになりつつあり、それらの蛋白質およびその遺伝子を精力的に解析することが重要な課題となる。特に日本で発見された複製開始のライセンス因子の解析は重要である。一方ARSの解析が進んでいる出芽酵母、分裂酵母の染色体を使ったin vivoでの研究も進める。研究項目B「染色体DNA鎖合成装置の分子構築」の分野では、複製フォークの進行に働く蛋白群の精製と再構築を目指して研究を行っていく方針である。研究項目C「染色体分配装置のダイナミクス」の分野では、原核細胞の染色体分配および細胞分裂の制御機構について種々の研究成果が話された。今後の方針としては、真核細胞の染色体分配機構の研究者にも多数加わってもらって研究を発展させていく計画である。
