ヒトセントロメア蛋白質CENP-Aの機能解析

研究概要	ヒト細胞染色体におけるセントロメア領域の機能が高頻度反復配列(αサテライトDNA)と一連のセントロメア蛋白質(コアヒストン、CENP-A、CENP-B等)との複合体を基礎とする繰返し構造にある可能性を追求している。本研究計画においては特にCENP-Aの機能解析に焦点を絞り解析を行っている。最近CENP-A遺伝子がクローン化されその塩基配列からヒストンH3に類似であることが予想された。我々はHeLa細胞由来ヒストンH2A、H2B、H3、H4およびCENP-Aを高純度に精製しヌクレオソーム構造再構成系を確率し、CENP-AがヒストンH3としてのヌクレオソーム形成活性を持つことを示した(論文準備中)。故にCENP-Aは遺伝子構造及び機能的にセントロメア特異的ヒストンH3であるとう事ができる。出芽酵母の系では染色体の分離分配に必須なヒストンH3類似の遺伝子(CSE4)が単離された。これらの知見はセントロメア領域にセントロメア機能を規定する特異的なヌクレオソーム構造の存在を示唆している。以上の知見を基礎に本年度はDNAと蛋白質との相互作用の観点から解析を行うために必須であるCENP-A抗体の作製を行った。大腸菌を用いた融合蛋白発現系によりCENP-Aタンパク質を発現し精製した。CENP-Aペプチドを数種類マウスマウスに免疫し、そのうちのいくつかがCENP-A抗体を産生することを確認した。今後モノクローン抗体を作製する予定である。
