研究概要

蛋白質の寿命決定の基盤となる選択的な蛋白分解の分子機構を明らかにするため、主要な蛋白分解系であるカルパイン、プロテアソーム系を中心に解析した。鈴木はヒトカルパインの力ルシウム非結合型の結晶構造を明らかにした。その結果、カルパインは活性を持たない酵素前駆体で、活性中心の形成には力ルシウムの結合によって誘発される構造変化が必須であることを明らかにした。また、ドメインIIIがカルシウム・リン脂質結合ドメインとして知られるC2ドメインであることを発見し、このドメインの力ルシウム依存的な構造変化がカルパインの活性化に必須の役割を持つことを推定し、実証に取りかかっている。牧はカルパスタチンの遺伝子を解析し34個のエキソンを同定した。プロモーターを異にする4種類のアイソフォームが存在する以外に、選択的スプライシングによる亜型の存在を確認した。田中はノックアウトマウスの作成から、プロテアソーム活性化因子PA28-γが個体成長に関与し、ポリユビキチン受容体遺伝子の欠損が卵の分裂異常によって胎生致死になることを明らかにした。また、γ-インターフェロンの刺激で26Sプロテアソームの調節因子PA700とPA28をもつハイブリッド型分子種が誘導され、内在性抗原のプロセッシング酵素として作用することを示した。山尾は分裂酵母のE2酵素を解析し、G2期の制御、分裂中期から後期への染色体分離およびM期のサイクリン分解ではAPCと共にubcP4が作用する以外に、別のE2、ubcP1を含む分解系の存在を示した。村上はオルニチン脱炭酸酵素(ODC)がアンチザイムとATP依存的に26Sプロテアソームで構造が解け、GroELシャペロンに捕捉されることを証明し、26SプロテアソームがODCのアンフォーディングを触媒することを示した。木戸は20SプロテアソームのC5、C8サブユニットがnucleotide diphosphate kinaseの活性を持ち、分子シャペロンとして機能することを推定した。
