研究概要	4つの重点項目について以下の研究成果を得た。(1)細胞周期制御:横沢は出芽酵母の増殖に伴って、定常期で顕著に26Sプロテアソームの活性と蛋白質量が変動することを見出し、増殖依存性の因子が26Sプロテアソームの構築を制御しているということを示唆した。岸本は細胞周期におけるM期の終了はCdc2-サイクリンB複合体のサイクリンBのAPCによる蛋白分解によってもたらされるが、Cdc2はその直前に解離し、APC以外の巨大分子複合体中に取り込まれていることを示した。(2)細胞増殖・分化の制御:名取はセンチニクバエ幼虫の体液細胞が、異物と接触した時に放出する二つのサブユニットからなるプロテアーゼは、一つの前駆体蛋白から翻訳後にプロセッシングを受けて生ずることを明らかにした。川島は下垂体由来GH4C1細胞において、TRHおよびTPA刺激によりカルパスタチンmRNA量の増加を観察し、PKCを介するカルパスタチンmRNA発現制御機構の存在を示唆した。(3)細胞死制御:三浦はショウジョウバエ複眼及びS2細胞においてCED-4の過剰発現は、caspaseの活性化を伴う細胞死を誘導すること、この活性化にはCED-4にあるP-loopモチーフが必要であることを示した。内山は、無血清、NGF無添加で誘起されるアポトーシスの系で、カスパーゼが活性化されない条件では、カテプシンD/Bによる細胞死制御系が働くことを明確に示した。(4)恒常性の維持・監視の制御:大隅は自食作用に必須な新しいタンパク質結合反応が関与していることを明らかにした。すなわちApg12pはそのC-末端Gly残基が標的タンパクApg5pのLys残基にイソペプチド結合を形成する。この反応は広く真核細胞に保存されている。木南はこの系にApg7pがE1様酵素として働いていることを明らかにした。また、神経性セロイド様リポフスチン蓄積症におけるプロテオリピッドのリソソーム内蓄積は新しいプロテアーゼの欠損によりおこることを示した。
