研究概要

カテプシンEおよびDは動物細胞における主要な細胞内アスパラギン酸プロテアーゼである。カテプシンDが典型的な可溶性リソソーム酵素として広く組織・細胞に分布しているのに対し、カテプシンEは非リソソーム酵素として限定的な分布をし、組織・細胞の違いによって細胞質型または細胞膜型、あるいはその両方の存在様式をとる。両酵素は明らかに糖蛋白質であり、その前駆体はシグナルペプチド、プロ部を有していることから、粗面小胞体上のリボソームで合成された後、古典的分泌経路でゴルジ野に運ばれるものと考えられる。しかし、両酵素、とくにカテプシンEの選別・輸送および活性分子への変換機構については不明の部分が多い。本研究では、マウス白血病フレンド細胞やラット胸腺細胞、ラットミクログリア細胞などの種やタイプの異なる細胞を用いて、カテプシンEの生合成や細胞内輸送ならびに局在化機構をカテプシンDのそれらと対比させて解析した。実験は、^<35>S-メチオニンを用いたパルス・チェイス実験により行われた。経時的に細胞と培養上清を回収し、細胞成分は界面活性剤で抽出した後、両酵素に対する特異抗体で免疫沈降を行い、ゲル電気泳動とフルオログラフィーで解析した。得られた結果は以下のとうりである。(1)カテプシンDはいずれの細胞においても同じキネティックスで生合成とプロセシングが行われ、数時間のうちにリソソームへ輸送され活性型酵素へ変換された。一方、(2)カテプシンEの生合成や細胞内輸送ならびに局在化機構は用いた細胞で著しい相違を示した。即ち、ミクログリア細胞では、カテプシンDと同様のキネティックスで生合成とプロセシングを受け、成熟型酵素として酸性コンパートメントへ輸送されたが、フレンド細胞や胸腺細胞では、中間型酵素の段階までしかプロセシングが行われず、この分子はトロンスゴルジあるいはTGNまで輸送されてプールされる形で貯留した。
