上丘一眼及び頭部運動系における視覚入力から運動出力への座標変換機構

研究概要

本研究では、前庭-頸運動系の研究をさらに発展させ、注視における視覚入力座標系から頭部運動系への座標変換の中枢神経機構を解析するため上丘から頸筋運動ニューロンへ運動指令情報を伝える視蓋脊髄路について電気生理学的、及び形態学的解析を行った。実験は、麻酔したネコで行い、上丘中間層から、深層へ電極を進めながら、電気刺激を行い、眼球運動のmotor mapを作製した。上方・下方・水平方向に大きな眼球運動を起こす部位を選んで、刺激電極を固定した。頸椎のC1,C2を開き、前索でHRPを充填した細胞内記録電極を軸索内で刺入し、対側上丘の電気刺激に応じてスパイクを生ずる軸索を捜し、上丘から直接スパイクが誘発された軸索と、単シナプス性に誘発されるスパイクを生じる軸索を選び、HRPを電気泳動的に注入した。頸筋の運動細胞を逆行性にラベルする目的で、頸筋の運動神経幹にHRPを注入し、DAB染色を行った。視蓋脊髄路細胞は、対側上丘の尾側2/3の中間層から深層に分布していた。前索を下行しながら、直角に次々と複数の軸索側枝を灰白質に出し、IX層に枝を出しながら、VIII,VII,時に,VI-V層の外側部に投射し、終末が見出された。この部は、トランスニューロナール染色により頚運動ニューロンに終わる最終オーダーの介在細胞の分布と一致した。単一細胞内染色によって上丘入力を受ける脊髄細胞も、この部に同定され、同側の前索を下行しながら多数の側枝を灰白質に出しており、神経終末は、VII-IX層、とくにIX層に多数存在し、逆行性に染められた複数の頚筋運動ニューロンの近位樹状突起にシナップス結合していた。以上により、上丘から頚筋運動ニューロンへの経路は、脊髄介在細胞を介して2シナップス性結合するのが主経路であること、又、単一介在細胞が多数の側枝によって、複数の頚筋を支配していることが明らかとなった。それが、単一前庭脊髄路細胞によって支配されるfunctional synergyのグループと同一であるか否かを今後明らかにする。
