研究概要

本研究班は、薬剤耐性マラリアを克服できる新しいマラリア治療薬の開発を行い、マラリア制圧に寄与することを研究目的としている。以下に研究実績を概説する。
1)薬剤耐性マラリア原虫のジヒドロ葉酸還元酵素(DHFR)の耐性機構を結晶学的に解析し、独自に開発した医薬品分子設計プログラムGREENを運用してマラリア原虫に選択的かつ耐性克服機能を付加した阻害剤(IMMD704)を設計した。IMMD704はマラリアのDHFRを選択的に強く阻害し、高い選択性を保持しつつ培養熱帯熱マラリア原虫を強力に阻害した。
2)抗マラリア作用を持つ化合物の検索:マラリア原虫の新しい培養系の確立と薬効スクリーニングの実施を行った。本重点領域研究班員、及び国公立、並びに民間の研究機関(45研究グループ)より抗マラリア作用検定の依頼のあった化合物(平成10年2月までに2321検体の評価を終えた)の薬効判定を行った。その結果、87検体に100倍以上の選択毒性を示す抗マラリア作用を認めた。特記すべきことは新規物質Febrinone Aに既存の抗マラリア剤と匹敵する抗マラリア作用(IC_<50>=1.8×10^<-9>M)を見出した。このFebrinone Aは次世代の抗マラリア剤と成りうる可能性がある。
3)マラリア原虫に特有な核酸代謝系を標的とした新しい治療薬の分子設計:
アンチセンス分子のマラリア原虫への適応を行い効果的な抗マラリア作用を示す標的分子の評価に使用出来ることを明らかにした。化学療法の標的としてのマラリア原虫ミトコンドリアの性質と分子生理学的な特徴を追求すると共に呼吸鎖電子伝達系の解明を行い、抗マラリア剤の標的となる酵素の構造解析を行った。さらに、マラリア原虫の陽イオン輸送性ATPaseは構造解析が終了し、液胞型H^+-ATPaseの解析を行っている。
4)ヒト型マラリア感染モデルマウスの作成を開始した。
