研究概要

(I)cDNAライブラリーの作成
1.Dictyostelium discoideum AX4株を用い、発現遺伝子種がもっとも多い移動体期の細胞より調製したpoly(A)+RNAに対してcDNAを合成し、size fractionationの後pBluescript IIへdirectional cloningすることによりライブラリーを作成した。
3.ライブラリーの一部を用い、インサートの長さ、cfu、クローンの重複の程度について検討し、ゲノム解析を行うに充分なqualityをもつライブラリーであることを確認した。
(i)cfu/6x10E4
(ii)インサートの長さの推定(平均長0.9kb)
移動体細胞より調整したpoly(A)+RNAに対するcDNAの1st strandをprobeにして行ったコロニー・ハイブリダイゼーション実験の結果は,85%以上が弱または中程度の強さのシグナルを示した。
(iii)一部クローンを塩基配列を決定し、98%がpolyA tailをもつRNA由来であること、また72%のクローンは全長または比較的長い塩基配列を含むことが分かった。
(iv)重複クローンの確率はおよそ10%であると推定された。
4.さらに、短いcDNAおよび長いcDNA用に、それぞれ0.5-1kbと1-21kbののcDNAを用いたライブラリーを作成した。
(II)データベースおよびホームページを構築し、インターネットで公開した。
また、細胞性粘菌・研究者の情報ネットにこれをリンクさせた。

(III)20000クローンの高密度グリッド化
(IIII)国際的なアッピ-ルと協力関係の樹立
細胞性粘菌の研究者のネットワークを通じて、日本が初の本格的なプロジェクトを開始したことをアッピ-ルした。また、仙台で開催された国際会議Dicty96では、ゲノムプロジェクトに関するワークショップを開いた。英国の2グループとは、我がグループが作成するcDNAの高密度化フィルターを用いて、細胞分化・形態形成異常の突然変異株を利用して、遺伝子の発現ネットを解明するプロジェクトを推進することにした。
