研究概要

アゲハはミカン、カラタチなどミカン科植物の葉を食べて生育するが、メス成虫はこれらの食草を前肢感覚毛により数種類の化学物質で認識して産卵行動を起こす。今年度、サンショウの葉に含まれる5'-p-coumaroylquinic acid,epicatechin,hesperidin,hyperinが産卵刺激活性を示すことを実証した。
チップレコーディング法を用いて、メス成虫前肢感覚毛の産卵刺激物質の電気的応答を検定した。その結果、食草植物抽出液に対しては小さい活動電位の反応がみられ、非食草植物に対しては大きな活動電位が生じ、明らかに応答の差がみられた。そこで前肢感覚毛は食草物質に対する特異的認識を示すことが判明した。個々の産卵刺激物質に対する応答はひきつづき研究中である。
アゲハ前肢感覚毛に存在するミカン葉産卵刺激物質に特異的なレセプターを探索する試みがなされた。アゲハ500羽から前肢末端を切断、RNAを抽出、PCR法で増幅した。メス画分に多く、オス画分に少ない成分をdifferential display法で分画していくつかのcDNA断片を分析した。その1つはP450-チトクロムbオキシーダーゼと判明したが、レセプターcDNAはまだみつかっていない。
アゲハの腹部末端にはオスにもメスにも光受容細胞が存在する。オスの光受容細胞はメスとの交尾に必須なことがわかった。メスの光受容細胞は交尾には必要ではないが、産卵には必要である。腹部末端から肛門突起を突き出させて葉をさぐりながら卵を産むさいに関与するものと思われる。
ラットの嗅上皮細胞にgreen fluorescence protein(匂いレセプターの一種)をアデノウイルスベクター法によって過剰に発現させることに始めて成功した。それらオクチルアルデヒドに対して電気的に反応した。
