研究概要	本研究は平成9年度から発足する重点領域研究「生体機能と設計分子」の準備研究として行われた。この重点研究は、情報伝達に関連した生体分子を標的として設計した探索分子を合成し、これを活用して生細胞および生体における標的分子の生理機能と病態への関与を統合的に解析・制御することを目的としている。この研究目的の成就には、新しい化合物の創製を可能にする有機化学および試験管内から細胞・個体にいたるまでの生体機能を総合的に評価する生化学・分子生物学・細胞生物学など異分野の研究者からなる研究チームの結成が必要である。この基盤研究(B)は、重点領域研究における上述したような背景にもとづき、研究分担者間での活発な討論を進めるべく申請される。この重点領域研究の準備段階として、班会議を平成8年10月18日と平成9年1月28日の2回行った。この重点領域研究で取り上げるべき標的分子や細胞現象を吟味し、各々についてどのような設計分子を合成・評価するかを以下のような項目について具体的に討議された。・蛋白質リン酸化による細胞内情報伝達系の修飾(機能探索分子によるプロテインキナーゼカスケードの解析・蛋白質リン酸化による膜リン脂質情報変換) ・受容体を介した情報伝達機構の分子解析 (プロスタグラジン受容体による情報伝達機構の研究・G蛋白質共役受容体の発現系の開発とその創薬への応用) ・情報分子の可視化による生理機能解析(Ca^<2+>イメージングによる細胞内Ca^<2+>局在と発生過程の解析・Ca^<2+>依存性開口放出の解析評価法) ・細胞情報探索分子の精密設計(合成プロスタグランジンの分子設計と合成・プロテインキナーゼC活性化制御のための特異的探索分子の設計及び合成) これらの項目は重点領域研究「生体機能と設計分子」において展開される機能探索分子設計の核になるべきものであり、これらの班会議を通して重点研究で挑発すべき研究テーマの準備は整っている。
