陸上植物の初期進化

研究概要	1.昨年度は、陸上植物の系統関係を色素体ゲノム上の5種類の遺伝子、4563塩基対のデーターを基に推定した。しかし、系統樹の解像度は1000塩基程度の配列を用いた場合と較べ、予想したほど良くならなかった。この理由は、色素体遺伝子の進化速度が目的とする系統関係を求めるには早すぎるためではないかと考えられる。そこで本年度からは、色素体ゲノムより進化速度の遅い、ミトコンドリアゲノム上の遺伝子であるCOX3遺伝子の塩基配列を用いて系統推定を開始した。これまで、苔類を中心に約10種の塩基配列データーが得られた。来年度、さらに他の分類群の配列を決定することにより、陸上植物の系統関係を明らかにする。 2.維管束分化、器官形成において重要な働きをする頂端分裂組織分化に関わる遺伝子群の候補を探すため、ミズワラビ、ニセツリガネゴケから、被子植物における研究より形態形成に重要な働きをしていることが知られているMADS遺伝子群、HD-ZIP遺伝子群、KNOX遺伝子群に属する遺伝子のcDNAを単離し、塩基配列を決定した。これらの遺伝子の発現様式、遺伝子系統樹から、各遺伝子と陸上植物形態進化との関連について解析を進行中である。昨年度作成した、ヒャクニチソウ維管束分化関連遺伝子であるTED3のプロモーター領域にレポーター遺伝子を融合したプラスミドをニセツリガネゴケに導入して、維管束が未分化であるニセツリガネゴケにおける発現様式から維管束進化のヒントを得る実験を進行中である。本年度は、効率良く、ニセツリガネゴケに形質転換をする技術導入を行い、安定して形質転換が行えるようになったので、来年度に向けて、形質転換体の形態観察を行っている。
