膵癌の発生・増殖と進展及びその制御に関する基礎的研究

研究概要

ハムスター膵癌実験系を用いた実験結果および進行状況について報告する。
1.膵癌の化学予防物質を検索する目的で、合成レチノイドである4-hydroxyphenyl retinamide(4-HPR)、Citrus coumalinの1種であるauraptene、天然カロチノイドの1種であるlicopene、非ステロイド系抗炎症剤であるpiroxicamのハムスター膵管発癌抑制効果について検索した。ハムスター短期膵管癌発生系において前癌病変の発生する時期より50日間上記の被検物質を投与した結果、0.02%および0.1%auraptene投与群において膵管病変数の発生が有意に減少したが、他の物質には有意な膵発癌抑制効果はみらえなかった。
2.食品中に含まれる発癌物質であるヘテロサイクリックアミンの膵発癌性について検索した。ハムスター短期膵管癌発生系において前癌病変の発生する時期より50日間被検物質として0.04%PhIP,0.06%MeIQx,0.03%IQ,0.02%Trp-P-1,0.02%Trp-P-2,0.08%AaC,0.06%DiMeIx,0.08%MeAaCを投与した結果、Trp-P-1およびDiMeIQxに膵発癌促進作用がみられた。
3.膵癌細胞で上昇しているテロメラーゼを標的とした膵癌の発生予防、治療法の確立のため膵癌細胞におけるテロメラーゼ阻害剤を検索した。ハムスター膵癌細胞株より蛋白を抽出し被検物質を混じてインキュベートしTRAP解析にてテロメラーゼ活性の阻害効果をみた。抗癌剤、抗生物質、抗炎症剤、天然化合物、抗酸化剤など約100種類の化合物を検索したが、テロメラーゼ特異的な阻害剤はみいだせなかった。
4.進行状況;ハムスターにおけるTrp-P-1の発癌性試験、膵癌細胞のアポトーシス誘導能を指標とした膵癌化学予防物質のin vitroスクリー二ング系の開発を計画している。
