膵癌の発生・増殖と進展及びその制御に関する基礎的研究

研究概要

ハムスター膵癌実験系を用いた実験結果および進行状況について報告する。
1. 短期膵発癌系を用いて、4-hydroxyphenylretinamide(4-HPR)の発癌修飾作用について検索した結果、0.0015%4-HPR投与群において動物1匹あたりの膵管病変発生数の有意な減少がみられた。一方、0.01%および0.04%4-HPR投与群において肝内胆管癌の発生頻度、動物1匹あたりの発生個数の有意な増加がみられた。
2. ハムスター膵癌において、アポトーシスに関与するwaf 1,bcl 2,bax遺伝子の発現を検索した結果、bax遺伝子の過剰発現がみられ、現在、この遺伝子の発現とアポトーシスの誘導、発癌過程における役割について検索を進めている。
3. 膵管上皮細胞のBrdU標識率を指標として、Trp-P-1の膵管上皮に対するイニシエイション活性を検索した結果、100mg/kgのTrp-P-1の胃内投与により膵管上皮細胞のBrdU標識率の有意な増加がみられ、Trp-P-1は膵管イニシエイション作用と膵発癌促進作用を有する膵のcomplete carcinogenであることが示唆された。
4. ハムスター膵癌において、matrix metalloproteinases(MMPs)の発現を検索した結果、膵管上皮過形成より浸潤癌へと進展するにつれMMP-2の発現が亢進すること、および浸潤癌においては活性型MMP-2の発現が増加することが判明した。また、短期膵発癌系において膵管上皮過形成の発生した時期より、MMPs阻害剤であるOPB-3206を飼料に0.1%および0.4%の濃度で混じ投与した結果、膵管癌の発生数の有意な減少がみられた。以上の結果より、膵癌の発生および進展にMMP-2が重要であることが示唆された。
