B細胞の分化,活性化,細胞死を制御する細胞内分子機構の解明

研究概要	自己赤血球に対する自己抗体をコードするH鎖遺伝子、L鎖遺伝子を導入した抗自己赤血抗体産生性トランスジェニックマウスでは、自己赤血球反応性B1細胞は体内に存在するが、無反応状態(アナジー)にある。このマウスをLyn欠損マウスと掛け合わせて、Lyn欠損抗自己赤血球抗体産生性トランスジェニックマウスを作成した。このような動物において、B1細胞における無反応性(または自己寛容)は破れて強力な自己抗体産生が引き起こされ、動物は重症の自己免疫性溶血性貧血に陥った。以上の結果は、LynキナーゼがBCRからのシグナル伝達において強い負の制御を行っており、Lynキナーゼによる抗原に対する閾値の制御は自己反応性B細胞の自己寛容維持に重要な役割を果たしている事が示唆された。マウスB細胞株WEHI-231を抗IgM抗体で架橋すると24時間以降にアポトーシスに陥る。この場合、カスパーゼの活性化とDNAの切断が誘導及び細胞膜透過性の亢進を引き起こす。カスパーゼ阻害剤Z-VADは核の分画化、DNA切断を完全に阻止するが、細胞膜透過性の亢進は阻止しない。WEHI-231細胞ではBCR刺激後、膜透過性の亢進に先立って、ミトコンドリア膜電位の変化および活性酸素の増加が生ずることを見出した。即ち、BCRからのDeath signalはまずミトコンドリアに伝達され、その後にカスパーゼの活性化とDNA切断及び膜透過性の亢進が生ずると考えられた。即ち、自己反応性未熟B細胞では、抗原受容体からのシグナルはまずミトコンドリアを標的して伝達され、その後にアポトーシス及びネクローシスの反応が引き起こされると考えられた。
