研究概要

我々はヒトDNaseIが遺伝的形質であることを発見した。法医鑑識科学や親子鑑定などの法医実務にDNaseI型を利用して多大な成果をあげることができ、DNaseI型がこの分野に利用できる極めて優秀な遺伝マーカーのひとつであることを証明した。従前の研究から、DNaseI多型について4対立遺伝子が同定されていたが、今回5番目の対立遺伝子(アリル)を発見した。アリル5はアリル2の1227ヌクレオチドにおけるG→A置換に基づくものであった。これに関連して、従来のDNaseI型判定法に使用したIEF-PAGE法に改良を加え、再現性良くすべての表現型を分離できるシステムを開発した。その結果、DNaseI型は15種類の表現型に分類でき、個人識別能の向上がもたらされた。さらに、日本人集団以外の民族集団にもDNaseI多型の存在が確認され、DNaseI多型は"world-wide"なものであることが明らかとなった。また、汗および汗付着靴下に関するDNaseI型の法医学的応用の結果から、微量な体液班からの個人識別マーカーとしてのDNaseI型の優秀性が再評価された。従って、DNaseI型は轢き逃げ車両に付着した微量な組織片から容疑者を特定する際に、極めて個人識別能に優れた遺伝マーカーであることが判明した。我々は、DNaseIIについてもその遺伝的多型の存在を報告しているが、本研究ではその完全長cDNA構造を解明した。これは脊椎動物DNaseIIについて初めて報告されたcDNA構造であった。さらに、轢き逃げ車両に付着した微量な組織片からの個人識別検査にDNA多型を導入するための基礎的知見として、2種類のSTRマーカー(HumTPOとHumLPL)について集団遺伝学的調査を実施したところ、個人識別において両STRマーカーの同時型判定が有効であることも明らかにした。
