研究概要

CAGリピート病の治療法開発を目的として,歯状核赤核・淡蒼球ルイ体萎縮症(dentatorubral - pallidoluysian atrophy, DRPLA)を対象疾患として,培養細胞を用いた様々な長さの野生型および変異DRPLAタンパクの過剰発現系を確立した。その結果,伸長したポリグルタミンのあるいはその周辺のアミノ酸を含む部分タンパク断片をCOS7細胞に発現させると,細胞内の核周囲および核内に凝集体の形成が観察された。この現象は全長の野生型DRPLAタンパクあるいはその断片では全く観察されなかった。この凝集体を形成した細胞は高率にTUNEL反応陽性となり,アポトーシスを生じていることが示された。このような現象は全長の変異DRPLAでは観察されないことから,変異DRPLAタンパクのプロセッシングにより生じる断片が重要な役割を果しているのではないかと考えられる。一方,DRPLA剖検脳においても小脳歯状核の神経細胞に核内封入体が存在することが見いだされ,神経細胞の核内凝集体形成が病態機序の上で重要であると考えられた。凝集体形成の機序については,ポリグルタミンがβ-シート構造をとりpolar zipperを形成するあるいはトランスグルタミナーゼが関与するとの説があるが,本研究で確立した培養細胞における発現系を用いてトランスグルタミナーゼ阻害剤を用いたところ,凝集体形成,アポトーシスが有意に抑制され,ポリグルタミンの細胞障害性を緩和できることを示した。この結果から、凝集体形成にトランスグルタミナーゼ反応が関与する可能性が示され,その活性を阻害することが治療法開発につながる可能性を示した。
