研究概要	本研究はヒトの皮膚メラノサイトにおけるメラニン形成過程とその生理的機能及び色素異常症(尋常性白斑)の分子生物学的病態の機序の解明を目的とする。その研究の実際的アプローチとしてチロシナーゼとチロシナーゼ関連蛋白(tyrosinase related protein 1;TRP-1)のcDNA co-transfectionの手技を用い解明する。このアプローチは従来の生化学的生物学的方法とは全く異なり、分子生物学的面からの新しい情報を得ることが期待できる。具体的には本研究において以下の5点を研究した。 1. チロシナーゼ遺伝子蛋白のfoldingと分子チャぺロン(カルネキシン)との関連 2. チロシナーゼ、TRP-1遺伝子の標的シグナルの解明 3. チロシナーゼ遺伝子蛋白とりソゾーム関連蛋白との分子レベルでの相互関係 4. 尋常性白斑のメラノサイトのメラニン形成異常の分子生物学的解明 5. メラニン形成と色素脱失機構の解明これらの研究によりメラノソーム生合成機序における初期メラノソーム(メラノソームstage I)が後期エンドソームに関係すること、チロシナーゼ遺伝子蛋白の輸送には小胞輸送が重要な働きを示し、この中にRab 7,PI-3 kinase等が加わること、またチロシナーゼ遺伝子蛋白の小胞体内での糖化におけるfoldingに際してはカルネキシンが重要な役割を占めることが分かった。また、メラニン形成の基質であるチロシナーゼを化学的に修飾し、実際にメラノサイトの選択的障害、及び色素脱失を起こさせることに成功した。得られた所見は尋常性白斑の病態の解明のみならず、他の色素異常症の新しい治療法の開発にも応用し得る。
