骨組織の加齢変化ならびに老化制御・再建機構に関する形態学的・分子細胞生物学的研究

研究概要

(1)メカニカルストレス実験:加齢および卵巣摘出によるラット下顎頭の変化
顎間接が加齢変化に伴ったメカニカルストレスによる影響を明らかにする目的で、様々な週齢の正常ラットおよび閉経モデルである卵巣摘出ラットの顎間接を骨形態測定学的に解析した。その結果、下顎頭は加齢変化に伴い骨量が増加し骨髄腔が狭窄する傾向を示したが、エストロゲンによる全身的調節を主に受ける下顎頭後方部とメカニカルストレスによる局所的調節が優位な下顎頭前方部に分けられることが明らかになった。以上、加齢変化に伴い、全身性ホルモンとメカニカルストレスの両方により下顎頭全体の骨構造の維持改変が行われていることが明らかとなった。
(2)骨誘導・骨形成促進実験:BMP-2,TGF-βに関する研究
骨形成誘導因子であるBMP-2を歯槽骨の抜歯窩に局所投与したところ、有意的に多量の新生骨が形成され、また間葉系細胞の増殖亢進および骨芽細胞への分化促進が認められた。さらに、骨形成誘導に関するTGF-β・・型レセプターの遺伝子発現および局所を検索すると、長管骨の骨端板軟骨の増殖細胞層下層から肥大化細胞層に認められ、また骨内膜の間葉系細胞にTGF-β・・型レセプターの局在が検出された。
(3)インプラント実験:
卵巣摘出によりエストロゲン欠乏ラットを作製し、骨組織に純チタンインプラント植立後の周囲骨形成および骨結合を検索したところ、卵巣摘出群ではチタン表面に形成される骨基質は薄く、骨芽細胞のマーカーであるアルカリホスファターゼ活性の低下および破骨細胞数の増加による骨吸収亢進が認められた。従って、閉経後の患者にインプラントを植立する場合、歯槽骨の骨代謝状態を考慮する必要性が考えられた。
