歯周病発症における病原性プロテアーゼの役割と作用機序

研究概要

歯周病の主たる病態は歯肉上皮ならびに結合組織などの比較的表層に存在する歯周組織の破壊とそれに引き続いて起こる歯槽骨の破壊である。こうした歯周組織の破壊には口腔細菌ならびに宿主細胞の産生するプロテアーゼが重要な役割を担っていると考えられている。しかし、歯周病の発症や進行過程におけるこれらのプロテアーゼの機能の詳細については依然として不明な点が多い。本年度の研究では、主要な歯周病原性細菌Porphyromonas gingivalisが産生する2つのユニークなシステインプロテアーゼArg-gingipain(RGP)およびLys-gingipain(KGP)の歯周病における機能と作用機序について遺伝学的ならびに生化学的な解析を行った。具体的にはRGP完全欠損株、KGP完全欠損株、RGP-KGP完全欠損株を作成し、野生株ももつ病原性の変化について生化学的な解析を行った。その結果、前年度の研究で示唆されていた以下の事実が確認された。すなわち、 (1)RGPは本菌の保有する好中球障害活性や血球凝集素活性の大部分を担っていること、 (2)RGPは線毛の主要成分であるフィンブリリンや75kDa表層蛋白質、さらにproKGPやproRGPなどの分泌蛋白質のプロセシングや細胞内輸送に関与していること、 (3)KGPは本菌の生長・増殖に必須のヘモグロビンの菌体への結合やヘミンの取込みに重要な役割を担っていること、 (4)KGPはフィブリンをよく分解し、炎症局所の出血傾向に寄与していること、 (5)RGPおよびKGPはともに強力な炎症惹起作用があり、歯周局所の炎症反応を担っていること、等々である。さらに、RGP・KGP完全欠損株を用いた研究から、両酵素が本菌の生長・増殖やエネルギー源として必要なアミノ酸をヘモグロビンなどの宿主蛋白質を分解して供給していることが示された。また、ヘモグロビンを結合するヘモグロビン受容体蛋白質の生成に両酵素が直接関与していることが明らかにされた。
