研究概要	研究代表者および研究分担者は、哺乳類細胞のヌクレオチド除去修復(NER)の分子機構解明を目指して,以下に述べる研究を行った。 1)色素性乾皮症(XP)C群蛋白質の機能解析 XPC蛋白質複合体のNERにおける役割を明かにするために、その構成成分の一つであるHHR23B蛋白質とその類縁蛋白質であるHHR23A蛋白質のXPC蛋白質との関係を調べた。その結果、HHR23A蛋白質もHHR23B蛋白質と同様に、XPC蛋白質と複合体を形成し、NERにおいて機能することを見出した。また精製蛋白質を用いた再構成NER系においても、HHR23B蛋白質およびHHR23A蛋白質がXPC蛋白質のNER活性を促進することを示した。 2)転写と修復のカップリングの解析 NERに必要な蛋白質因子(XPB、XPDなど)を含む基本転写因子TFIIHのコンポーネントの中で、p62及びXPBが他のNER蛋白質(XPGおよびXPC)と直接相互作用していることを明らかにした。 3)新たな修復因子の同定酵母の2ハイブリッドシステムを用い、HHR23B蛋白質と相互作用する因子を検索した。その結果、26Sプロテアソームの構成因子の一つであるs5a蛋白質が相互作用する因子として見出された。これら2つの蛋白質は、細胞抽出液中でも相互作用していた。 4)C群以外のXP蛋白質の精製と新たな修復因子の同定 XPF蛋白質とERCCI蛋白質とを昆虫細胞の系で共発現させ、活性のある形で精製することに成功した。
