研究概要	本研究者らは既に、中枢神経系である視床部位のPGI_2受容体に強く結合する受容体探索分子(15R-TIC)の開発に成功し、中枢神経系に特異的に発現する新規なプロスタサイクリン受容体の存在を証明している。本年度、研究分担者の細谷は15R-TICの構造修飾により、この受容体にさらに強く結合する探索分子の合成に成功した。また、その構造を基に受容体タンパク質捕獲のための新たな光親和性標識リガンドを設計し、その合成にも成功した。現在、この新規受容体の生理機能の解明を目的とした生化学実験が進行中である。一方、研究代表者の鈴木は、抗腫瘍性プロスタグランジンの人工類縁体であるΔ^7-PGA_1メチルエステルの血清中での代謝挙動を解析し、このPGが異性化酵素の作用によりΔ^7-PGC_1を経て迅速に代謝されることを明らかにした。さらに、この問題を克服すべく、代謝酵素との分子認識という観点からPGの立体化学に着目した分子設計を行い、代謝に安定でかつ活性および持続性の高い抗腫瘍性プロスタグランジンを創製することに成功した。現在、これらPGの細胞に対する作用のスクリーニングを行っている。一方、前年度に明らかにした癌細胞のPG耐性メカニズムの知見を基に、PG細胞外輸送に中心的役割を果たしているGS-Xポンプの競合的阻害剤を設計し、その合成に成功した。予備的ではあるが、この阻害剤を抗腫瘍性PGと併用することにより耐性癌細胞のPG感受性が上がることを確認している。また、共同研究者である根岸らは抗腫瘍性PGによるストレスタンパク質BiPの誘導作用を明らかにし、PGの細胞内標的タンパク質の検索を検討している。以上、本年度の研究は概ね計画通りに遂行できた。
