MMDA受容体チャネルの多様性と脳機能

研究概要

1)海馬CA3野錐体細胞の頂上樹状突起に存在する交連/連合繊維入力によるシナプスでは、NMDA受容体チャネルε1サブユニットの欠損によりNMDA受容体応答およびシナプス長期増強(LTP)が低下した。一方、基底樹状突起に存在するfimbrial(Fim)入力によるシナプスでは、ε2サブユニットを片方の染色体で欠失させたヘテロ接合マウスでNMDA受容体応答およびLTPが低下することを見い出した。すなわち、海馬CA3野錐体細胞は多様なNMDA受容体チャネルの分子種をシナプスにより使い分けていることが示唆された。2)NMDA受容体チャネルε1、ε2およびζ1サブユニットに対する特異抗体を作製し、組織切片の前処理による新たな免疫組織化学法を考案した。NMDA受容体チャネル依存性のシナプス可塑性が見い出される海馬CA1野の各層およびCA3野の分子層、放射層、上昇層にはNMDA受容体チャネルサブユニットが豊富に分布しているが、シナプス可塑性がNMDA受容体チャネル非依存性であるシナプスが集積するCA3野の透明層にはサブユニットの局在が乏しいことを明らかにし、錐体細胞ないでのNMDA受容体チャネルサブユニットの局在がシナプス特性を決定していることを示唆した。3)海馬を含む前脳からシナプス後肥厚(PSD)分画を単離し、カルシウムとカルモデュリンを投与して内在性のCaMKIIを活性化することにより、PSDに共存するAMPA受容体チャネルがリン酸化されることを明らかにした。このリン酸化は、CaMKIIに特異的な阻害ペプチドで著明に抑制されたが、他のリン酸化酵素の阻害剤ではほとんど抑制を受けなかった。この実験結果から、LTPにCaMKIIによるAMPA受容体チャネルのリン酸化が関与する可能性が示唆された。4)グルタミン酸受容体チャネルδ2サブユニット欠損は、小脳プルキニエ細胞の棘突起と平行線維終末との馬の結合率を選択的に低下させることを見い出した(結合率:野生型100%、ノックアウト63%)。発達段階において、このシナプス結合率の低下は生後第2週より顕在化した。δ2サブユニットは生後第1週まではプルキンエ細胞の細胞体や樹状突起に広く分布しているのに対して、生後第2週以降はシナプス局在が確立することから、δ2サブユニットは発達段階における平行線維シナプスの結合強化・安定化に関与することが明らかとなった。5)背根神経節神経の生長円錐伸展抑制分子として同定されたセマフォリンIIIの生体内での役割を明らかにするために欠損マウスを作成した。胎生12日の欠損マウスにおいて末梢神経の走行に異常を見い出した。すなわち、動眼神経以外の三叉神経、顔面神経、迷走神経の軸索走行が拡散し、肢芽の軟骨組織や眼球上皮への神経の異常な進入が認められた。これらの結果から、セマフォリンIIIが生体内で神経回路網の形成に関与していることが明らかとなった。
