Elongation法によるタンパク質のThrough Bond電子移動反応機構

研究概要	本年度は、これまでのElongation法にもとづくプログラムに、局所状態密度を求める部分を付け加えた。すなわち、タンパク質のモデル系として、10乃至30個のアミノ酸残基からなるオリゴペプチドを考え、その各アミノ酸残基上のHOMO付近、LUMO付近の局所状態密度を求めたところ、アミノ酸残基の配列の順序および相互の立体的な配置に大きく依存することが明らかになった。すなわち、タンパク質内において、アミノ酸配列やその相互の立体的配置により、各アミノ酸残基部分の電子供与性や電子受容性、さらには反応性が大きく異なることを示し、大変興味深い結果が得られた。さらに、Elongation法により、ポリペプチド鎖を伸長していく過程で、Through Bondによる効果もみることがわかった。つぎに、この結果を実際の3次元タンパク質に応用できるようにするために、階層的にポリペプチド鎖の電子状態を求めることを考えた。すなわち、まずある長さのオリゴペプチドの電子状態をElongation法で計算する。ついで、このElongation法によって高分子鎖の電子状態を求める過程で得られる局在化分子軌道のうちで、高分子鎖の末端部に存在するものをActive空間として、異なる二本の高分子鎖の電子状態を接続することを試みた。すなわち、上記の二本の高分子鎖のActive空間の局材化軌道を基底として、固有値問題を解くのである。モデル計算として、例えば10個のグリシン残基からなる2本のオリゴペプチドに、上記の方法を適用することにより、信頼性が高く計算時間の面でも大変有利であることがわかった。この方法により、まずある程度の長さのオリゴペプチドの電子状態をいくつか作り、それらを相互に結合することにより、タンパク質そのものの電子状態を階層構造的に理論的合成できることがわかった。
